Bud and Lou were the pet hyenas of Harley Quinn and the Joker, which Joker had bought at some point after the Christmas Eve event.
At some point in time after the Arkham Asylum Riots, they were shot and stuffed either by the Penguin or someone on his orders, after which being put on display in the Cyrus Pinkney Natural History Museum (Penguin's base) as a trophy in a victory against the Joker.
Riddler discovered their bodies, connecting a riddle to them and tasking Batman to solve it in order to capture him and locate the hostages he had imprisoned.
